Remember to adjust brightness and contrast of the image for best display. When creating the overlay of both images in FIJI remember to invert one image. 

The signal obtained by group A and B does not allow to do the quantification of all samples in Fiji. It's a real experiment! Mention in SLIMS, also for answers to Q3 and Q4. 

For group C and D: ELN submission extended to after the holidays. Note that quantification of bands in L+ and L- is sufficient. Quantify and compare the signal intensity of the loading control between both L+ and L- lanes. Then compare the signal intensity of recombinant AMY2-MYC-HIS between both L+ and L- lanes; normalize based on loading control. Mention in SLIMS when the data is lacking to answer questions, for instance for answers to Q3 and Q4. 

• Group A: 1° anti-6xHIS OR anti-myc-tag 500 ng/ ml; AND anti-actin 100 ng/ul
  
• Group B: 1° anti-6xHIS OR anti-myc-tag 1000 ng/ ml; AND anti-actin 200 ng/ul (different batch)
  
• Group C/D: 1° anti-myc-tag (one group anti-6xHIS) 1000 ng/ ml; AND anti-actin 1000 ng/ul
