Description

Determine transformation efficiency

  • Since all groups A had extremely low counts for plate C there was likely a mistake. The issue could have been the cells or the tube with the + control plasmid. I tested the transformation efficiency of cells with a fresh batch of + control plasmid, and their transformation efficiency was as expected (plate C hundred to thousand(s) of colonies)
  • For Group B, C, D the aliquots with + control have been adjusted
  • If plate C was too numerous to count you may use 1500 CFU for the calculation. Note, ideally one should have plated two different dilutions, for example 200 ul and 50 ul, and thus obtained a countable number for the lower dilution
  • For group C: download the zip folder, then unzip to access the images
  • For Group D: remember that you might have plated 50 ul on plate C (instead of 200 ul)
